How does melatonin affect other hormones?

Melatonin is itself a hormone, mainly known for signaling biological night. It also interacts with reproductive and other physiological pathways, but that does not mean a sleep supplement predictably disrupts every hormone. Effects depend on context, and long-term hormonal consequences—especially during development—remain incompletely understood.

Circadian signaling is its clearest role

Endotext describes melatonin’s daily light-dark rhythm and receptor-mediated actions, including effects on the body’s central clock. Taking an external dose changes exposure to that signal. This is different from supplying an essential nutrient or replacing every aspect of the natural nightly rhythm. [1]

Reproductive pathways are biologically connected

A 2025 review discusses effects through the hypothalamic-pituitary-gonadal axis and melatonin receptors. Much of that evidence concerns animal reproductive physiology and seasonal breeding. The authors identify uncertainty about the relationship in humans; animal findings cannot predict a fixed testosterone, estrogen or fertility response to a retail dose. [2]

Development deserves particular caution

NCCIH notes unresolved questions about longer-term use in children, including possible effects on puberty, menstrual cycles and prolactin. This uncertainty is not proof that all use causes disruption, but it supports pediatric oversight rather than assuming that a natural hormone is harmless at any age. [3]

Evaluate symptoms instead of self-balancing hormones

Imagine someone develops a persistent reproductive symptom while using a sleep supplement. Review the exact product, timing and other medicines with a clinician. Do not add another hormone product to counter a presumed mechanism without confirming the problem. [3][4]
