Dihydroberberine absorption: a five-person pilot did not show better glucose control

A short crossover experiment found higher berberine exposure with a derivative. Glucose and insulin did not significantly differ, so the exposure result is not proof of superior treatment.

What the experiment compared

Five healthy men received placebo, berberine or two dihydroberberine regimens in a crossover experiment. Four doses preceded a two-hour blood-sampling period with a test meal. [1]

Some exposure comparisons favored dihydroberberine, but baseline levels differed and several comparisons were only trends. Glucose and insulin showed no significant differences. [1]

Interpretation and limits

An investigator disclosed paid advisory work for NNB Nutrition under an institutional conflict-management plan; funding was not established from the abstract. The tiny sample, brief observation and healthy population cannot establish long-term metabolic superiority or a personal dose conversion. [1]

A hypothetical distinction

Imagine one preparation raises a blood concentration more than another while the clinical marker does not change. Higher exposure remains an absorption finding, not a demonstrated clinical advantage.
