Melatonin and serotonin: connected chemistry does not make them interchangeable

Serotonin is an intermediate in the body’s melatonin synthesis pathway. That does not mean taking melatonin reliably raises brain serotonin, treats depression or reproduces the effects of a serotonin-targeting medicine.

The pineal synthesis pathway has a direction

Endotext describes tryptophan-derived serotonin being converted to N-acetylserotonin and then melatonin. Enzymes and timing signals regulate that process. A diagram showing serotonin upstream of melatonin is not evidence that swallowing the final product drives the pathway backward and increases serotonin. [1]

Melatonin provides information about biological night

NHLBI describes melatonin release during darkness as a signal helping prepare for sleep. Light exposure and the body clock shape the rhythm. Sleep regulation involves more than a single molecule, so a simple more-serotonin-equals-more-sleep rule is misleading. [2][1]

A biochemical connection is not a medication substitution

The inspected physiology source does not establish melatonin as a serotonin-raising treatment. Do not replace prescribed antidepressants or combine sleep products based on that inference. NHS advises reviewing antidepressants and other medicines before melatonin because interactions and sedation can matter. [1][3]

Read a pathway without turning it into a dose plan

Imagine a diagram places serotonin before melatonin. It explains how one substance can be used to make another, not which supplement a person needs or what will happen to brain levels after a pill. Clinical questions require evidence about the actual exposure and outcome. [1]
